12mm stone is significant. It won't go off with medication. It can increase in size with time. It can cause recurrent episodes of pain and infection. These kind of stones can be cleared by procedure called RIRS. In this flexible ureteroscope is passed upto kidney. And stones are fragmented with laser. No scars on body. No risk of bleeding. One day stay in hospital.
