Cotton you can after 5-10 minutes or as soon as clotting is done. Pain will also diminish in few days and so the mark.

The cotton can be removed once the bleeding stops, usually within 1-2 minutes... for patients on blood thinning medications it might take more time.... for that a bandaid can be applied... and the pain usually subsides in 24 hours
